Phenotype
Homozygous knock-in mice (SRBI G319V/G319V) suffered >95% lethality by weaning. There is elevation of total cholesterol and LDL-C in SRBI G319V/wt (heterozygous) mice compared with littermate controls. After 4 weeks of a modified Western diet (21% fat, 1.25% cholesterol), significantly elevated HDL-C in female SRBI G319V/wt mice compared with littermate controls and a trend toward elevated HDL-C in heterozygous male mice was observed.

Strain Development
CRISPR guide(s) and Cas9 protein were microinjected into C57BL/6J zygotes and progeny were screened for the desired mutation via sanger sequencing. Founders were mated to C57BL/6J breeders (Jackson Laboratory #000664), and derived N1 progeny were identified by sequencing. N1 were then mated again to C57BL/6 breeders to generate N2 mice identified by sequencing. N2 heterozygous mutant mice were mated for production of phenotyping cohorts. Due to embryonic lethality, similar to SR-BI knockouts, these mice were later backcrossed five generations into the 129S1/SvImJ (Jackson Laboratory #002448), although this did not improve lethality. Genotypes are all confirmed via PCR and sequencing.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
