Genetic Alterations
Naa10R83C knock-in (KI) mice harbor a CRISPR/Cas9-induced R83C missense variant and a silent R82R mutation in exon 5 of the N(alpha)-acetyltransferase 10, NatA catalyctic subunit (Naa10) gene. The Naa10 gene is part of a major N(alpha)-acetyltransferase complex responsible for alpha-acetylation of proteins and peptides. Mutations in this gene have been associated with developmental delay, attention-deficit/hyperactive disorder (ADHD) like behavior and cardiac abnormalities.

Phenotype
Heterozygous mice appear phenotypically normal and are fertile. Homozygous females and hemizygous males are sub-viable and born below expected Mendelian ratios (20.9% and 17.9%, respectively, from Het x Hemi matings). Homozygous females and hemizygous males are fertile but are occasionally non-productive in breeding. Hemizygous males are smaller in size with lower body weight than WT littermate males, but no differences are observed in heterozygous or homozygous females.
Strain Development
The Naa10R83C knock-in (KI) allele was generated using CRISPR/Cas9 endonuclease-mediated genome editing. A guide RNA (TCTGAGCCAGGCCAAGGCGC), and a single stranded oligo donor carrying the missense R83C (CGC>TGC, arginine to cysteine) variant and a silent R82R (CGG>AGA) mutation, were used for HDR-mediated genome editing. The R83C missense and R82R silent mutations are in exon 5 of the N(alpha)-acetyltransferase 10, NatA catalytic subunit (Naa10) gene on chromosome X. Mouse Naa10-201 transcript (ENSMUST00000033763.15) was used as reference for the exon number and guide sequences. Guide RNA, cas9, and the ss oligo donor were introduced to single cell C57BL/6J zygotes and transferred to pseudo pregnant females.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
