Genetic Alterations
A targeting vector was generated so that a DTR-EGFP-pA-loxP-neo-loxP cassette (diphtheria toxin (DT) cDNA linked in-frame to EGFP polyA-floxed neo) was inserted immediately after the ATG codon in exon 1 of the Lgr5 locus (deleting the remainder of exon 1 and splice donor site in intron 1 - a 212 bp deletion) by homologous recombination in C2 ES cells. Correctly targeted clones were transfected with a cre plasmid to remove the neo cassette.

Phenotype
Heterozygous mice are fertile and healthy, with no observed phenotypes. The model allows labeling and/or ablation of Lgr5+ (GPR49+Gpr49+) intestinal stem cells. The allele is homozygous lethal due to lack of LGR5 protein (Lgr5 KO phenotype).

Strain Development
A 7,213 bp fragment from a C57BL/6 BAC (RP23 library) was first retrieved into plasmid pBlight-TK25. To generate the DTR–EGFP KI vector for Lgr5, a DTR–EGFP-pA-loxP-Neo-loxP cassette was synthesized and inserted right after the ATG of Lgr5, deleting the remainder of exon 1 and splice donor of intron 1 (a 212 bp deletion). To generate the CreERT2 KI vector, a dsRed2-IRES-CreERT2-pA-Frt-neo-Frt cassette was synthesized and inserted at the same position as the DTR–EGFP cassette. The Lgr5 KI vectors were linearized with NotI. C57BL/6 C2 embryonic stem cells were targeted using standard methods. Positive clones were identified using PCR and taqman analysis, and confirmed by sequencing of the modified locus. Correctly targeted embryonic stem cells were transfected with a Cre to remove the Neo cassette. The modified embryonic stem cells were then injected into blastocysts and germline transmission was obtained after crossing the resulting chimaeras with C57BL/6N females. The colony has been maintained by intercrossing and backcrossing (F23 and N15) to B6 albino (Strain #:000058).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
