Genetic Alterations
C57BL/6J mice were microinjected with CRISPR/Cas9 genome editing technology and a LOX-STOP-LOX (LSL) cassette. This cassette replaced the first intron 180 bp upstream exon 2 in the Nfe2l2 gene and included an E79Q mutation inside exon 2.

    Phenotype

    Nullizygous mice have increased sensitivity to oxidative stress in organs and cells, abnormal tooth enamel, altered response to injuries/chemical treatments, age-related decline in spermatogenesis and ovarian follicle numbers

    Conditional phenotype: expression of the mutant Nfe2l2-E79Q allele is silent until exposed to Cre recombinase. Activation of the Nfe2l2-E79Q allele in all K14-positive alleles driven by a K14-Cre allele. This resulted in hyperplasia/hyperkeratosis in the epithelium of the forestomach, esophagus and oral cavity. Other phenotypes observed included: stunted body axis, decreased weight, and decreased visceral adipose depots.

    Strain Development
    A donor vector comprised a 2.3 kb segment of the mouse Nfe2l2 gene encompassing exons 2 and 3, including the E79Q mutation in exon 2, several silent mutations to remove PAM sites, and a LoxP-flanked stop cassette. This cassette was microinjected with CRISPR/Cas9 reagents into C57BL/6J embryos, where homologous recombination with the donor vector occurred in the endogenous Nfe2l2 gene.Pups were screened to identify founder animals harboring the correct knock-in allele. A male founder with the knock-in allele was mated to wild-type C57BL/6J females to produce F1 heterozygous pups.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
